I would like to know if it would be possible to exchange the start and end points, once the application has designed the flight.

With normal missions there is no way to do that but with linear missions you can do it using the rotate gesture. For normal missions you will have to manually rotate the mission to where you want it.

Yes, all you need to do is rotate the mission 180 degrees to get that effect.
