Unlock mission to modify after drone is in the air

How can I unlock a mission once it has started and the drone is in the air?  I see how I can pause the mission, but after that I want to fly to precise points and move my mission markers to that point.  I don't see the "unlock" after the mission has started, and when I upload even after unlocking, the mission reverts to locked.  Thanks.
